I guess I can... but since it's so late I'll put up a tough one and see if it lasts all night...

Thanos was once the owner of all 6 infinity gems, my question is:

Whom did he acquire them from and how?

I never read any of that story, I don't think...
    so I'll guess.

from Death, and by promising to end all life in the universe.

    sounds Thanos-ish enough.

Stupid work keeping me occupied all day...

Toymank is correct, they were taken from Elders...

And Peeker, you're answer is also correct, but in a sense of, that was "Why" he did.

I'll list the how down below... if anyone is interested, this all takes place within the pages of Thanos Quest 1 & 2.

Soul gem: Thanos departs on his mystical chair and heads for the nexus of realities, a place at the border between the realms of Chaos & Order. He traverses different realities and arrives. There, the In-Betweener sits in a sphere imprisoned. The gem Thanos seeks is the soul gem. It affects the soul of a person and can alter their very spiritual being. He convinces the In-Betweener that he has escaped Death's servitude and is seeking protection from her. He surmises that the In-Betweener's magic combined with his power might be able to break the sphere/prison surrounding the In-Betweener. Feeling he has nothing to lose, the In-Betweener agrees to attempt it and they both focus enormous proportions of energy at the sphere. The prison gives and the In-Betweener is free. Having no loyalty, the In-Betweener tries to destroy Thanos but soon realizes he's powerless. Thanos explains that they are in a realm in between Chaos and Order's universes and that he is powerless there. Inside the sphere/prison, the In-Betweener retained power only because his masters allowed it for his convenience and comfort. Thanos then approaches him and removes the gem from his forehead. He tells the In-Betweener that he is one of those beings who exists outside the realm of destiny's sphere of influence and that Chaos & Order cannot see/detect him. He leaves the In-Betweener as Chaos & Order appear angry at the escape attempt. A scream is heard as the In-Betweener futile tries to explain to his masters...

Power gem: Thanos arrives at a planet where he sees Champion (the Elder) fighting/battling the natives - something Champion lives for. The gem Thanos now seeks is the power gem. A gem that can be tapped into for brute strength or can be used to power the other gems. Thanos challenges Champion to a fight with the planet they are on as a prize. They begin fighting but Thanos easily outwits his opponent. Attempting to anger him, Thanos soon erects a force field around himself. Champion attempts to break it but just gets angrier. He then jumps into the air wishing to fall down and break the force field with a concentrated blow. As he falls down, concentrating all his power on the punch, Thanos at a precise moment teleports away and Champions' power punch shatters the entire planet by causing inner core chain reactions. Champion thinks he's won as he hangs on to a piece of rock from the planet that is floating in space, but Thanos appears on his mystical chair and tells him he hasn't. That all he has done is destroy the prize of their contest. Champion challenges Thanos but Thanos turns him down and begins to leave. Champion realizes he is stranded and asks Thanos to not leave him there. Thanos tells him it will cost him his gem. Though he resists at first, Thanos reminds him that though he is immortal, he may well be there several centuries. Champion agrees and gives Thanos his gem. Thanos tows him to a nearby planet and then lets him fall from orbit as he decrees that he'd take him to the closest planet but never agreed to a soft landing...

Time Gem: Thanos arrives and begins talking to the Gardener, an Elder dedicated to his beautiful garden. A most beautiful sight which even Thanos acknowledges . Here he seeks the time gem. A gem capable of manipulating time such that you can travel within the time stream as easily as you can walk, or freeze time to prevent any states of changes within an object. The two civilly discuss the Gardener's membership as an Elder. The Gardener replies he only became an Elder to get the gem which he wanted for his garden to improve and retain its beauty. He claimed he did not like the Elder group as they were too power hungry for his liking. Thanos offers the Gardener a chance to relinquish his gem, but the Gardener claims he must be true to his nature despite what may happen. Thanos looks down with regret as vines begin to encircle him. His eyes glow as he explains that the Gardener subconsciously used the gem's power to accelerate the growth of the plants and as they bloomed froze them in time to keep their beauty. He then expresses regret as he reveals that he used the power gem to feed the time gem causing exponential sporadic growth. The Gardener is seen dead with plant limbs protruding from his body and orifices. Thanos then leaves with three gems in his possession...

Space gem: The Space gem Thanos seeks can transport you anywhere, everywhere and nowhere instantly. It has the capacity to warp space to take the holder anywhere they wish. In the blink of an eye, the Runner destroys Thanos' mystical chair and quickly places Thanos at his mercy. He then dictates terms to Thanos of revealing all he knows and why he is after the gems or getting beat to death at super speed. Thanos reveals that gazing at the Infinity well, he learned of a being that existed before recorded time. It was a single lone entity with limitless power existing within any and all realities. It then chose to put an end to itself of which Thanos had surmised was due to loneliness. From its ashes arose all that is reality and the core became six infinity gems. The Runner then tells Thanos the gems are now his but Thanos stops feigning defeat and uses his time gem to age the Runner over a million years old. He tells the Elder that they are not truly immortal - just extremely decelerated metabolisms. Thanos then takes the space gem and regresses the Runner to a child form. He then teleports with the Runner baby inside the stronghold of the Collector...

Reality gem: The Reality gem Thanos seeks to gain from a trade can control what you see, feel and sense through your perceptions as well as what is. The Collector is initially shocked at Thanos offering his very own Elder to his collection. He agrees with the condition that Thanos promise to non-interference in his existence. Thanos promises and the exchange is made. A sampling of the reality gem takes place as the room is distorted out of reality. The Elder realizes he's been duped and asks Thanos to leave. Before he does, Thanos reveals that the effects of the time spell on the Runner are temporary and the Runner subsequently quickly changes from the baby form back to his normal self. At the realization that the Collector was going to add him to his collection, the Runner begins pummeling him incessantly. The Grandmaster turns off his screen after observing the interchange between Thanos and the Collector. He realizes where Thanos is headed to next...

Mind gem: The last gem - the mind gem, Thanos seeks next. It is the gem that can penetrate and control the mind. Telepathy and telekinetics are added to your skills with this gem. Thanos teleports to a planet and quickly requests to the Grandmaster what the game will be. The Grandmaster begins by explaining that the gem he has is protected by layers of force fields and that a pan-dimensional transporter will teleport the gem to a random location in the universe immediately and then self destruct if any layer is breached leaving no way to trace. He then continues on by stating the game to be a quasi military exercise on a mental plane. Each contestant is equipped with an identical suit of battle armor. A "kill shot" immobilizes the loser and terminates the game. The Grandmaster requests Thanos to barter his five gems and Thanos agrees since he doesn't plan on losing. The Grandmaster tells Thanos the transporter will activate should anything interrupt his vital signs and that on the other hand, it will shut down in the event he loses. Thanos accepts the terms and places his gems on a stasis field keyed to his vital signs. The game begins as they both place their headsets on. Thanos finds himself in a battle suit inside a maze-like arena. The Grandmaster quickly finds Thanos and begins firing but Thanos is able to dodge. He goes for cover and then tries to fire back but the Grandmaster has gone. Taking to the air, the Grandmaster assaults an unexpecting Thanos firing from above. Thanos runs for cover again but this time blasts a hole and hides within the maze wall. The Grandmaster flies by leaving Thanos his back. The Grandmaster realizes his mistake. As he turns around, Thanos tries to fire but is instead covered like a statue by a silicon fungus that will eat through him. The Grandmaster tells him he loathed cheating but that the stakes were too high. Gloating, the Grandmaster knocks the covered statue down only to discover it was a robot. On the surface of the planet, the real Thanos destroys the mental plane machine leaving the Grandmaster trapped inside. Thanos then obtains the mind gem exclaiming, "Out of a thousand ways I could have taken the 6th gem, this one was surely the most enjoyable one!"

So it's Toymank's question... since he was the closet...
